Workshop Description

This workshop will cover the basis of Next-Gen Sequencing Library Preparation for Illumina Sequencers. Different Library Preparation Techniques (DNA-seq, ChIP-seq, RNA-seq, Methyl-seq) are explained the first and second day in class. The third day, Quality Control steps of the starting input material and final libraries are performed in the Lab (TapeStation). Purification from Primer Dimers (using AMPure beads) and Library Submission is also performed on the same day. The third day is optional, sign-up is required.

  • Theory behind Next Generation Sequencing Library Preparation
  • DNA Library Preps (WGS and WES)
  • Other Library Preps:
    • DNA Methylation
    • ChIP-seq
    • ATAC-seq
  • RNA Library Preps:
    • mRNAseq
    • total RNA
    • small RNA
    • single-cell seq

(Pellegrini Lab – TLSB, room 4021)

  • Quality control of RNA samples
  • Quality control of final libraries
  • AMPure beads Purification and Size Selection of Sonicated DNA

Technical Requirements

Day 2 will take place in a wet lab. Attendees will need the following prior to the start of class:

Dr. Marco Morselli is currently a post-doc in the Lab of Professor Matteo Pellegrini. He received his bachelor and master degrees in Molecular Biotechnology at the University of Parma, Italy. He earned his PhD at UCLA focusing on the crosstalk between histone modifications and DNA methylation. Outside of the lab, Marco enjoys cooking, running, cycling and playing soccer.

It was a super clear lecture. I learned a lot and also was very entertained the whole time. Marco answered questions great and obviously prepared very well for this. Thanks to The Collaboratory for making this happen.

Clear and informative!

Fall 2018 Student
In combination, the lectures and the hands on experience were invaluable in understanding the core concepts of library prep. Thank you for the opportunity to attend the workshop and update my knowledge. Keep up the great work!
Fall 2018 Student
Keep up with the good work! Very information-, action-, and humor(quiz/game)-packed short course.
